Lion’s Mane vs Bacopa: An Honest Comparison

This is one of the most common questions I get, and for a long time I answered it badly. The standard version presents two memory supplements and asks which is better — but after going carefully back through both literatures, I don’t think that’s quite the right question. These two compounds have fundamentally different types of supporting evidence, and both are marketed on a promise — a dramatic memory boost — that neither really delivers at the advertised scale.

So this article does something slightly different: it compares the quality and shape of the evidence rather than the marketing claims, then gives a clear recommendation anyway. Why the usual comparison is misleading

Both compounds are sold primarily on memory. Bacopa carries a long Ayurvedic history as a memory remedy; Lion’s Mane is marketed on nerve growth factor and the implication of a brain that physically rebuilds itself. Two products, same promised benefit, so a head-to-head seems natural.

The difficulty is that the evidence underneath is shaped completely differently in each case. Bacopa has been tested repeatedly in humans, and those trials show modest cognitive effects — some genuine memory free-recall gains, plus faster speed of attention — a real but small signal that the “3,000-year memory herb” marketing wildly oversells on magnitude. Lion’s Mane has barely been tested in healthy humans at all, so its memory claim rests almost entirely on mechanism. Comparing them head-to-head on memory therefore means weighing a compound whose human effect is demonstrated-but-small against one whose human effect is largely undemonstrated. That’s not a like-for-like contest, and it’s why most versions of this article reach confident conclusions they can’t justify.

Bacopa: more human data, modest benefit

Bacopa’s advantage is straightforward: it has been studied in humans more than most botanical nootropics. A systematic review identified six randomised controlled trials, all running 12 weeks, using standardised extracts at 300–450mg daily, and found that Bacopa improved performance on 9 of 17 tests in the domain of memory free recall, with little evidence of enhancement in other cognitive domains (Pase et al., 2012). So the human trials do show a memory effect — specifically in free recall — which is worth stating plainly, because it’s genuine.

The catch is magnitude. When the trial data were pooled meta-analytically across 437 participants, the most robust effect was on speed of attention — a shortened Trail B time of about 18 milliseconds and reduced choice reaction time — leading the authors to conclude Bacopa may benefit cognition particularly in the attention domain (Kongkeaw et al., 2014). And a well-known trial in healthy older adults found Bacopa significantly improved delayed word recall and Stroop performance versus placebo, while also reducing anxiety and depression scores (Calabrese et al., 2008).

Put together, Bacopa’s honest profile is a modest, repeatedly-tested compound with real — if small — effects that span memory free-recall, speed of attention, and possibly anxiety. That’s less dramatic than the packaging, and an 18-millisecond gain on a lab task is not the transformation the marketing implies. But it’s grounded in more human data than almost anything else in this category, and the effects it does show are the kind you’d have to measure to notice, not feel.

Lion’s Mane: better mechanism, thinner evidence

Lion’s Mane has the more compelling story and the weaker human record. Its compounds — hericenones and erinacines — can induce nerve growth factor synthesis in nerve cells (Lai et al., 2013), with further work showing neuronal survival and neurite outgrowth via the TrkA/Erk1/2 pathway (Zhang et al., 2017). Both are studies in cultured cells. Among its active compounds, erinacine A has confirmed pharmacological action in the central nervous system in rats (Li et al., 2018). It’s an elegant, coherent mechanism — genuinely more interesting than Bacopa’s.

The human side, though, rests almost entirely on one trial: 30 Japanese adults aged 50–80 with mild cognitive impairment, randomised into two groups of 15, taking 3,000mg daily for 16 weeks, with improved cognitive scores at weeks 8, 12 and 16 — and scores falling significantly four weeks after they stopped (Mori et al., 2009). That’s fifteen people per arm, in a clinical population, in an industry-affiliated study. Researchers reviewing the field have themselves described it as still some way from bench-to-bedside translation (Brandalise et al., 2023). For a healthy adult seeking enhancement, that’s close to no directly relevant human evidence at all.

The real choice: which uncertainty do you prefer?

Framed properly, this isn’t a contest between a stronger and a weaker compound. It’s a choice between two different kinds of not-knowing.

With Bacopa, you have repeated human testing and a modest, somewhat unglamorous result. The uncertainty is about magnitude — the effects are real but small, and whether you’d notice an 18-millisecond speed gain or a modest free-recall improvement in daily life is genuinely open. With Lion’s Mane, you have an elegant mechanism and almost no human data in healthy people. The uncertainty is about existence — whether the effect is there at all in someone like you.

If you prefer betting on something small and demonstrated, Bacopa. If you prefer betting on a plausible mechanism that may prove larger once properly studied, Lion’s Mane. My own view is that for someone new to this, the demonstrated-but-small option is the better use of money and attention — which is why Bacopa appears in our beginners guide and Lion’s Mane doesn’t.

If memory is genuinely your goal, buy neither first

Most people arrive at this comparison because they want to remember things better — names, study material, what they read. If that’s you, the honest answer is that neither capsule is where the leverage is. Bacopa’s memory effect is real but small; Lion’s Mane’s is largely unproven in healthy people. Picking one and stopping there would be doing you a disservice.

Two interventions have dramatically better evidence than either compound. 🧮 Worked example — testing one of these without fooling yourself

The single biggest self-testing mistake here is tracking the wrong outcome. People take Bacopa expecting a dramatic memory lift, don’t feel one, and quit — while the effect the trials actually found (a small free-recall gain, faster reaction speed) sails past unmeasured. Here’s how to run a test that could actually detect what’s there:

1. One compound at a time. Never start both together — each needs 12+ weeks to judge, so running them at once guarantees you learn nothing about either. Pick one and commit to a single 12-week block.

2. Choose measures that match the evidence. For Bacopa, track the things its trials actually moved: a timed reaction-speed task and a short delayed-recall test (e.g. learn 15 words, recall after 15 minutes) — and, since it showed up in the data, a simple weekly anxiety self-rating. Don’t judge it solely on “do I feel sharper.”

3. Baseline for two weeks first. Run those measures several times before dosing, to capture your normal range and its natural wobble. A single “before” number tells you nothing about whether a later change is real.

Worked judgement: at week 12, compare your dosing scores to the baseline range, not a single low day. If your reaction speed or delayed-recall sits consistently above that range, that’s a signal worth keeping. If it’s inside your normal variation — the most likely outcome, honestly — the fair conclusion is “no detectable effect for me,” and the right move is to stop. I take both, which makes me a poor advertisement for choosing between them — but the way I take them differs, and that difference is the honest summary of this whole article. Bacopa I take because a body of human trials points at a small real effect, and because when I look back across my own notes the pattern that holds up is slightly lower baseline agitation during heavy work periods. Lion’s Mane I take because the mechanism is elegant and the safety profile is excellent, and I have never been able to detect a thing.

Neither has moved my Creyos scores in any way I’d defend. That’s not a claim that they do nothing — my n=1 testing isn’t powerful enough to detect an 18-millisecond change or a modest free-recall gain, and I shouldn’t pretend otherwise. It’s a claim that I can’t personally verify what the trials suggest, which is a different and more honest statement than most people in this space make about their own stacks.

If someone asked me to strip back to one, I’d keep Bacopa — more human data, and the calm-during-busy-periods observation is the closest thing to a real signal I’ve got from either. And if they were choosing their first-ever supplement for memory specifically, I’d tell them to spend the money on nothing and fix their sleep instead. That advice has never once made me a commission, and it’s still the correct advice.

For both compounds, what matters is knowing exactly what’s in the capsule. Bacopa should state its bacoside standardisation, because that’s what the trials used. Lion’s Mane should state fruiting-body content or a declared dual extract, because the category is poorly standardised and grain-substrate padding is a real problem. Third-party testing on both.

Bacopa has the better memory case, but it’s modest. Which works faster?

Both are slow, and neither will give you anything noticeable in the first few weeks. Every Bacopa trial in the systematic review ran 12 weeks (Pase et al., 2012). In the Lion’s Mane trial, significant differences from placebo first appeared at week 8 and continued through weeks 12 and 16 (Mori et al., 2009). Practically, a fair test of either requires a three-month commitment. If you want something you can assess the same day, L-theanine with caffeine is a far better first experiment than either compound here.

Which is safer?

Both have reassuring profiles with different specific cautions. Bacopa is generally well tolerated, with gastrointestinal effects the most commonly reported issue — largely preventable by taking it with food — though it warrants caution alongside thyroid or anticholinergic medication. Lion’s Mane is a culinary mushroom eaten as food across East Asia, and its 16-week trial reported no adverse effects on laboratory tests, but anyone with a mushroom or mould allergy should avoid it, and respiratory and skin reactions have occasionally been reported. Long-term data is limited for both.
